engine guys?

if i accidently set my timing with the engine on the oppisite stroke (intake or exhaust) would it still run? i am having a pain in the ass time figuring out whats wrong with my truck. i built a new engine put it in and it will crank after playing with it for a while. but when i shut it off, i have to play with the timing for about 30 minutes for it to run again.
